METHODS Circulating tumor cells were detected in preoperative blood samples using reverse transcription polymerase chain reaction (RT-PCR) for alpha-fetoprotein (AFP) mRNA. Midkine expression was immunohistochemically examined in surgically resected HCC tissues. RESULTS Forty-one HCC patients with positive serum AFP were studied. Positive AFP mRNA expression was shown in 18 preoperative blood samples (43.9%) and midkine overexpression was detected in 29 excised tumors (70.7%). Among the 29 patients, 18 (62.1%) showed positive AFP mRNA in their preoperative blood samples. In contrast, the positive AFP mRNA expression was only observed in 3 out of 12 patients (25.0%) without aberrant midkine expression. Statistical analysis showed that abnormal midkine expression in the tumors was a variant significantly associat&~ with the presence of tumor cells in the blood circulation (P<0.05). CONCLUSION In human hepatocellular carcinoma, midkine overexpression is correlated with a haematogenuous spread of the primary tumor cells, and ~t may be a marker for tumor metastatic potential.

Status of serum alpha feto-protein (AFP) and midkine (MDK) levels in patients with hepatocellular carcinoma
Introduction: Hepatocellular carcinoma (HCC) is sixth common cancers worldwide and predominant in Asia and Africa. A number of evidence suggests a possible role of midkine (MDK) and α-fetoprotein (AFP) in the pathogenesis of hepatocellular carcinoma (HCC). Materials and methods: We studied MDK and AFP in patients with HCC or in healthy controls. MDK and AFP was measured by enzyme linked immuno...
متن کاملmiR-26b enhances radiosensitivity of hepatocellular carcinoma cells by targeting EphA2
Objective(s): Although low-dose radiotherapy (RT) that involves low collateral damage is more suitable for hepatocellular carcinoma (HCC) than traditional high-dose RT, but to achieve satisfactory therapeutic effect with low-dose RT, it is necessary to sensitize HCC cells to irradiation. This study was aimed to determine whether radiosensitivity of HCC cells can be enhanced using miR-26b by tar...
متن کاملRad51 Expression in Nasopharyngeal Carcinoma and Its Association with Tumor Reduction: A Preliminary Study in Indonesia
Background: Overexpression of Rad51 protein in many tumor cells has been proven to increase radioresistance and can be related to the resistance of chemosensitivity of tumor cells. This preliminary study was conducted to determine the relationship between the Rad51 expression level in nasopharyngeal carcinoma and the response of the treatment based on the measurement o...
متن کاملMidkine promotes hepatocellular carcinoma metastasis by elevating anoikis resistance of circulating tumor cells
Midkine is overexpressed in hepatocellular carcinoma (HCC) and plays a role in tumor progression, but less is known about its role in resistance of circulating tumor cells (CTCs) to anoikis which leading to recurrence and metastasis. The aim of the present study was to analyze whether midkine was associated with HCC progression with anoikis resistance. We found that cultured HCC cells were more...
متن کاملPrognostic Value of HLA-G in Malignant Liver and Pancreas Lesions
Background: Human leukocyte antigen (HLA)-G is a nonclassical HLA class I molecule with modulatory effects on NK and T cells. Because HLA-G expression is frequently detected in different solid tumors, it may be involved in tumor immune evasion. Objective: This study was designed to elucidate the prognostic value of HLA-G in hepatocellular carcinoma (HCC) and pancreatic adenocarcinoma (PADC). Th...
